Metabolisme van nier-endotheelcellen in gezondheid en acute nierschade: een opportuniteit voor nieuwe therapeutische mogelijkheden

Chronic kidney disease is a growing public health concern already affecting close to 10% of the European population. The only viable option for end-stage renal disease patients is kidney transplantation, yet the number of good quality donor kidneys does not meet up to the actual demand at all. Contributing to this is the ischemia/reperfusion injury (IRI) to which kidneys are unavoidably exposed during graft preservation and transplantation. Renal endothelial cells (RECs; cells lining blood vessels) are crucial in normal kidney function and are extremely sensitive to IRI yet virtually nothing is known about how their (metabolic) heterogeneity (per renal compartment) affects their response to IRI. With state-of-the-art transcriptomics and bioinformatics, I will interrogate metabolic alterations in REC subtypes during IRI with the overarching aim of identifying new metabolic targets that can be exploited to protect the renal vasculature from IRI. As a proof-ofprinciple for such metabolic intervention, I will further optimize the use of ketone bodies (alternative energy sources) and elucidate the exact molecular mechanism behind their protective effect in renal IRI, for which the host lab obtained preliminary but convincing data. I will have the opportunity to validate my (most promising) data in ex vivo perfused pig kidneys and by immunostaining human kidney sections. As such, this project has a high potential for fundamental science discovery and for clinical translation. status: published
